A procedure is described for the trapping and identification of hydrogen selenide and methyl selenol ( CH3SeH ). The volatile selenols were generated by reducing selenious acid or dimethyldiselenide with Zn dust and hydrochloric acid under a stream of nitrogen and passing into a trapping solution composed of 50 mM 1-fluoro-2,4-dinitrobenzene plus 83 mM sodium bicarbonate in 67% dimethylformamide:33% water. The selenols react rapidly to form stable dinitrophenyl (DNP) selenoethers that can be extracted into benzene; these are easily identified by TLC, HPLC, or mass spectrometry. Hydrogen selenide is trapped in 90-99% yield, primarily as the di-DNP- monoselenide with a trace of di-DNP- diselenide .  相似文献

Eleven citrus groves of diverse varieties and ages infected with Tylenchulus semipenetrans growing in differing soils in Florida were treated with three rates of 1,2-dibromo-3-chloropropane (DBCP) applied by various means. Yield, fruit size, and T. semipenetrans populations in the roots were compared between DBCP-treated and untreated trees over a period of I-3 yr. Maximum fruit size and yield were obtained by applying DBCP at 38-58 kg/hectare (ha) (34-52 lb/acre); whereas best nematode control was with a rate of 77 kg/ha (69 lb/acre). Application of chemical emulsion with a special, drilled, low-profile sprinkler irrigation ground pipe was the most suitable method. Effect of DBCP treatment generally lasted for 3 yr. A mean annual I. 1% increase in fruit diameter, 15.2% increase in fruit yield and a 55.7% decrease incitrus nematode populations was found for D BC P-treated trees in contrast to untreated trees.  相似文献

In a survey of antagonists of nematodes in 27 citrus groves, each with a history of Tylenchulus semipenetrans infestation, and 17 noncitrus habitats in Florida, approximately 24 species of microbial antagonists capable of attacking vermiform stages of Radopholus citrophilus were recovered. Eleven of these microbes and a species of Pasteuria also were observed attacking vermiform stages of T. semipenetrans. Verticillium chlamydosporium, Paecilomyces lilacinus, P. marquandii, Streptomyces sp., Arthrobotrys oligospora, and Dactylella ellipsospora were found infecting T. semipenetrans egg masses. Two species of nematophagous amoebae, five species of predatory nematodes, and 29 species of nematophagous arthropods also were detected. Nematode-trapping fungi and nematophagous arthropods were common inhabitants of citrus groves with a history of citrus nematode infestation; however, obligate parasites of nematodes were rare.  相似文献

1 Although the weaver ant Oecophylla is the first written record of biological control, dating from 304 ad , there have been fewer than 70 scientific publications on this predator as a biological control agent in Asia, from the early 1970s onwards, and fewer than 25 in Africa. 2 Apart from crop‐specific ecological and perceptual factors, a historical review shows that political and market forces have also determined the extent to which Oecophylla was incorporated into research and development programmes. 3 In Africa, research on weaver ants in biological control concentrated on export crops, such as coconut and cocoa, whereas, in Asia and Australia, research focused on fruit and nut crops, primarily destined for domestic markets. 4 Increased evidence of pesticide inefficiency under tropical smallholder conditions, changing paradigm shifts in participatory research and a growing scientific interest in local knowledge in the early 1990s opened up new avenues for research on conservation biological control. 5 Lobbying and advocacy have been needed to ensure that Oecophylla was recognized as an effective biological control agent. 6 With an increased market demand for organic produce, holistic approaches such as conservation biological control, particularly the use of Oecophylla, are increasing in importance. 7 Multi‐stakeholder strategies for collaborative learning are proposed for a better control of major fruit, nut and timber tree pests in Africa, Asia and Australia.  相似文献

The analysis of the volatiles released by the novel bacterial isolate Chitinophaga Fx7914 revealed the presence of ca. 200 compounds including different methyl esters. These esters comprise monomethyl‐ and dimethyl‐branched, saturated, and unsaturated fatty acid methyl esters that have not been described as bacterial volatiles before. More than 30 esters of medium C‐chain length were identified, which belong to five main classes, methyl (S)‐2‐methylalkanoates (class A), methyl (S)‐2,(ω?1)‐dimethylalkanoates (class B), methyl 2,(ω?2)‐dimethylalkanoates (class C), methyl (E)‐2‐methylalk‐2‐enoates (class D), and methyl (E)‐2,(ω?1)‐dimethylalk‐2‐enoates (class E). The structures of the compounds were verified by GC/MS analysis and synthesis of the target compounds as methyl (S)‐2‐methyloctanoate ( 28 ), methyl (S)‐2,7‐dimethyloctanoate ((S)‐ 43 ), methyl 2,6‐dimethyloctanoate ( 49 ), methyl (E)‐2‐methylnon‐2‐enoate ( 20a ), and methyl (E)‐2,7‐dimethyloct‐2‐enoate ( 41a ). Furthermore, the natural saturated 2‐methyl‐branched methyl esters showed (S)‐configuration as confirmed by GC/MS experiments using chiral phases. Additionally, the biosynthetic pathway leading to the methyl esters was investigated by feeding experiments with labeled precursors. The Me group at C(2) is introduced by propanoate incorporation, while the methyl ester is formed from the respective carboxylic acid by a methyltransferase using S‐adenosylmethionine (SAM).  相似文献

Volatile organic compounds (VOCs) are secondary metabolites acting as a language for the communication of plants with the environment. In orange fruits, the monoterpene D-limonene accumulates at very high levels in oil glands from the peel. Drastic down-regulation of D-limonene synthase gene expression in the peel of transgenic oranges harboring a D-limonene synthase transgene in antisense (AS) configuration altered the monoterpene profile in oil glands, mainly resulting in reduced accumulation of D-limonene. This led to fruit resistance against Penicillium digitatum (Pd), Xanthomonas citri subsp. citri (Xcc) and other specialized pathogens. Here, we analyze resistance to pathogens in independent AS and empty vector (EV) lines, which have low, medium or high D-limonene concentrations and show that the level of resistance is inversely related to the accumulation of D-limonene in orange peels, thus explaining the need of high D-limonene accumulation in mature oranges in nature for the efficient attraction of specialized microorganism frugivores.  相似文献

Abstract

Co-v-culture (co-cultivations of physically separated microbes that only interact through the air) systems were designed to investigate the effects of microbial volatile organic compounds (mVOCs) from about 20 different microbes, on a medicinal fungus, Ganoderma lucidum. For more accuracy in co-cultivations, a novel synchronized cultivation approach was tested for culturing G. lucidum. The hyphal growth of G. lucidum and the content of its ganoderic acids (GAs) were measured. In almost all of the co-v-cultures, there was an inhibiting effect on hyphal growth and a promoting effect on GAs contents. In inducing GAs production, Bacillus cereus PTCC 1247 and Pseudomonas aeruginosa UTMC 1404 were the most effective ones, as, compared to control cultures, GAs content increased 2.8 fold. Comparing different co-v-cultivations demonstrated that the concentrations of mVOCs, oxygen, and carbon dioxide were the main players in co-v-cultures. No correlation was found between hyphal growth and GAs production. Strains of the same species imposed totally different effects on hyphal growth or GAs production. This study has investigated the effects of mVOCs on G. lucidum for the first time. Moreover, it suggests that co-v-cultivation may be a promising biotechnological approach to improve the production in G. lucidum.  相似文献

Abstract

We have previously described a method to capture, identify and quantify volatile components in expired breath. The purpose of this research is to provide a non-invasive means to measure biomarkers of metabolism in vivo. In the present studies, the effect of 1-aminobenzotriazole (ABT), an inhibitor of diverse cytochrome P450 (P450) enzymes, on the composition of volatile organic chemicals (VOCs) expired in the breath of male F-344 rats was determined in parallel with the catalytic activities and total content of hepatic P450. lntraperitoneal administration of ABT (100 mg kg-1) to rats resulted in markedly diminished hepatic microsomal P450 content and activities. The extent of inhibition was near maximal at 4 h, at which time approximately 50% of the total P450 content, about 65% of the CYPlA2 activity, 55% of the CYP2E1 activity, and about 80% of CYP2B activity were lost. Inhibition was maintained to 48 h post-dosing, but P450 content and activities had largely been restored by day 7. Concomitant with the inhibition of P450 were corresponding increases (up to several hundred-fold) in the molar amount of volatiles appearing in the breath of ABT-treated animals, and the rebound of P450 levels was attended by corresponding decreases in the appearance of breath volatiles. These studies indicate that P450 plays a major role in the metabolism of VOCs appearing in breath, and that these chemicals can serve as markers on P450 activity in vivo.  相似文献
